What is Melanotan 1?

Melanotan 1 (MT1), also known as afamelanotide, is a synthetic analogue of the naturally occurring hormone alpha-melanocyte-stimulating hormone (α-MSH).

It was developed to investigate the regulation of skin pigmentation, melanin production and the biological response to ultraviolet radiation.

Melanotan 1 primarily activates the melanocortin-1 receptor (MC1R), which is expressed on melanocytes, the specialised cells responsible for producing melanin.

Activation of this receptor stimulates melanogenesis, the biological process through which melanin pigment is produced.

Increased eumelanin production has been investigated for its potential role in the skin's natural response to UV exposure.

Unlike Melanotan 2, which has broader melanocortin receptor activity, Melanotan 1 is more selective for pigmentation-related signalling.

A prescription implant formulation of afamelanotide, marketed as Scenesse, has been approved in certain countries for specific medical indications, including erythropoietic protoporphyria (EPP). This approval does not extend to unapproved MT1 research vials or cosmetic tanning products.

How It Works

  • Melanin Production: Activates MC1R signalling pathways associated with increased melanin synthesis in melanocytes

  • Skin Pigmentation Research: Investigated for its influence on eumelanin production and skin pigmentation

  • Photoprotection Pathways: Studied for its potential role in the biological response to UV-induced skin stress

  • Melanocortin Receptor Activation: Mimics aspects of naturally occurring α-MSH signalling, primarily through MC1R

  • Cellular Signalling: Activates cyclic AMP (cAMP) pathways involved in regulating pigmentation-related enzymes

  • Skin Biology Research: Investigated in studies examining pigmentation disorders, light sensitivity and melanocyte function

Increased pigmentation does not guarantee protection against UV damage or skin cancer. MT1 is not a substitute for sunscreen or other sun-protection measures.
